Can I stain both cell surface and intracellular targets simultaneously when preparing cells for mass cytometry?
Yes. The standard protocols for staining intracellular targets involve fixing followed by permeabilization of the cells, which allows probe access to both cell surface and intracellular targets. Some cellular target epitopes are altered by fixation and/or permeabilization, which can affect probe binding. In these cases, you can either optimize antibody clone and/or fix/permeabilization condition to enable detection of the problematic target, or stain surface markers prior to fixation, then fix/permeabilize and stain for intracellular markers.
