What is the difference between SNP Type Normalization and no template control (NTC) normalization?
SNP Type Normalization is the default normalization method for Fluidigm SNP Type assays. It helps guide automatic calling of genotypes even when nonspecific signal is observed in the NTC ("floating NTC"). SNP Type Normalization determines a global NTC position based on the majority position of NTCs across the entire IFC run.
NTC normalization is the default normalization method for TaqMan Assays. Each TaqMan SNP Genotyping assay contains two dually labeled probes, which may differ with respect to background fluorescence. NTC normalization takes into account these differences and attempts to position each NTC at a 0.1, 0.1 position on the scatter plot.
