The progesterone receptor (PR), also known as NR3C3 (nuclear receptor subfamily 3, group C, member 3), is a steroid receptor that binds progesterone. The steroid hormones and their receptors are involved in the regulation of eukaryotic gene expression and affect cellular proliferation and differentiation in target tissues. Human PR is expressed as 2 isoforms: the full-length PR-B and the short form, PR-A. Although the 2 isoforms have similar DNA- and ligand-binding affinities, they differ in transcriptional activities. Progesterone receptor isoform B (PR-B) is involved in activation of c-Src/MAPK signaling on hormone stimulation, while PR-A functions as a strong transdominant repressor of PR-B.
