Nestin is a class VI intermediate filament (IF) protein expressed in a variety of stem and progenitor cells, including primitive neuroepithelial cells and pancreatic exocrine progenitor cells. Structurally, nestin has the shortest head domain (N-terminus) and the longest tail domain (C-terminus) of all IF proteins. Nestin regulates not only protein function but gene function via direct binding to DNA. Nestin expression is used extensively as a marker to identify CNS stem cells in the developing nervous system and in cultured cells in vitro.
