Foxp3, a member of the Fox protein family of transcriptional regulators, is a master regulator of the regulatory T (Treg) CD4+ cell subset. CD4+CD25+Foxp3+ Tregs play a suppressive role within the immune system, and loss-of-function mutations of Foxp3 in the scurfy mouse leads to devastating widespread lesions. Foxp3 contributes to maintaining the suppressive phenotype of Tregs by acting as both a transcriptional activator and a repressor, including by increasing the expression of CD25 and CTLA-4 and by reducing expression of IL-7Rα and IL-17. Some of these effects are due to its binding to the proinflammatory Th17 master regulator ROR-γt.
