STAT6 is a transcription factor involved in response to the related cytokines IL-4 and IL-13 as well as other stimuli such as PDGF. Canonically, cytokine receptor engagement facilitates recruitment of STAT6 to members of the JAK family, which activate it by phosphorylation at Tyr641. Non-JAK-mediated activation has been reported downstream of viral infection, poly(I:C) treatment, and the oncogenic fusion protein BCR/ABL p190. Activated STAT6 can form homodimers or heterodimers with other STAT proteins, translocate to the nucleus, and bind DNA to facilitate transcription of target genes, including many involved in Th2 polarization.
