Monoclonal antibody 6B11 reacts with a unique determinant in the CDR3 region of the invariant (Vα24-JαQ) TCR chain. 6B11 identifies invariant NKT (iNKT) cells, which are a small subset of T lymphocytes that also express other NK cell molecules such as CD161. Vα24-JαQ TCR interacts with the glycolipid-loaded MHC class 1b molecule CD1d, inducing activation and subsequent cytokine production. CD1d-presented antigen results in rapid production of IL-4 and IFN (also known as IFN-γ). It has been reported that iNKT cells play an immunoregulatory role and participate in antitumor immunity and autoimmunity.
