CD244.2, also known as 2B4, is a 38 kDa type I transmembrane protein and member of the CD2 subset of the immunoglobulin superfamily of molecules. 2B4 is expressed on NK cells, monocytes, basophils, and eosinophils and is inducibly expressed on a subset of CD8+ T cells in both mice and humans. In NK cells, 2B4 has been reported to have both activating and inhibitory functions. However, evidence in both murine and human models indicates that its role in T cells is co-inhibitory. 2B4 expression is reduced in patients with systemic lupus erythematosus (SLE), and 2B4 deficiency in mice resulted in spontaneous development of a SLE-like disease in autoimmune-prone genetic backgrounds.
