The 621 monoclonal antibody reacts with human GITR (glucocorticoid-induced TNF receptor family-regulated gene), also known as AITR (activation-inducible TNFR family member), or CD357. GITR is an approximately 25 kDa member of the TNFR superfamily. Reports in the literature suggest that GITR signaling can regulate T cell proliferation and TCR-mediated apoptosis. GITR associates with TRAF1, TRAF2 and TRAF3 and induces nuclear factor NF-κB activation via TRAF2. GITR binds GITRL/AITRL to mediate the development of regulatory T cells and to regulate the activity of T cell Th1 subsets.
