Lymphocyte activation gene-3 (LAG-3) is a type I transmembrane protein expressed on activated CD4+ and CD8+ T cells, a subset of gamma delta T cells, NK cells, and regulatory T cells (Tregs). LAG-3 binds to MHC class II molecules with higher affinity than CD4, and the function of LAG-3 is dependent on this interaction. LAG-3-deficient mice do not exhibit any phenotypic defects. However, they undergo an increase in expansion compared to wild-type T cells in a lymphopenic environment. Both thymically and peripherally derived Tregs lacking LAG-3 are defective in their suppressive activity, and ectopic expression of LAG-3 imparts regulatory phenotype to CD4+CD25- T cells.
