CD47 is a 42-52 kDa member of the immunoglobulin (Ig) superfamily, which contains a single Ig variable region (IgV)-like extracellular domain and regulates multiple cellular processes implicated in immune responses. It is widely expressed on hematopoietic and non-hematopoietic cells. CD47 makes numerous protein-protein interactions with cell surface and soluble ligands including integrins, thrombospondin (TSP-1) and signal regulatory protein alpha (SIRPα). Recently, CD47 has been considered as a therapeutic target for acute myeloid leukemia (AML) since it was noticed that CD47 is expressed on AML cells at higher levels than on their normal counterparts, and that increased expression is associated with poor clinical outcomes.
